Some of my favorite missions feel dated now - in that they're pretty simple and there's only one or two ways to complete them - so it's a bit unfair to compare them to the newer games. So this list is in order of most challenging to least challenging (but still fun/memorable):

1. LaTouche
2. Lafrčniere
3. Lucy Thorne
4. Laurens Prins
5. Cesare Borgia (final mission in Viana)
6. Sibrand
7. William of Montferrat
8. Antonio Maffei
9. The Pope in AC2 (infiltrating the Vatican)
10. Al Mualim (including the eerie zombiefied Masyaf)

I'll have to mull this over now, but I'm interested that dimbismp had Prins as his number 2 spot. I mean, to each their own, but was I missing something with that mission?

Well, both Sixkeys and LoyalACFan also have this assassination in their top 10s.

I like this assassination because it has many layers. First you have to get through 3-4 groups of guards at the plantations. It's quite challenging to do so without getting detected. Then you get to the mansion. Again this place has lots of guards and you have to be careful and avoid rushing to the target. The white room confrontation is really cool and memorable:
-"[...]i was only after a little bit of coin"
-"as was I,as was I"

This dialogue shows to the player that at this point of the story,Edward is no different than the supposedly evil slavers etc. He would do anything for a little bit of coin.Finally,the chase after the assassination is also pretty good.

Finally,the fact that it is this mission that the real nature of James Kidd/Mary Read is revealed,is also a plus.

All of this, plus the fact that it's completely open-ended with several viable paths (i.e. across the rooftops, through the second-story hallways, around the backyard perimeter, etc).

Ok, cool!

Yeah, I'm not criticizing or anything, I just personally don't really like that mission, and didn't know it was apparently pretty popular. Good to know!
