So it says:
Three lives toiled.
Three lives lost.
Three messages delivered:
The greeting.
The warning.
The revelation.



What could this mean?
is the 3rd Connor or Desmond?

This makes sense And three means probably Altaïr, Ezio and Desmond.

Im pretty sure it all means TWCB.

Three lives toiled (Minerva, Juno & the bearded geezer whose name escapes me were all tasked to find a solution to save life)
Three lives lost (they failed)
And the three messages relate to the 3 meetings we've had with TWCB

3 lives toiled: Altair, Ezio, and Connor? We can't count Connor out. In a way they were prophets for Desmond, they delivered knowledge/messages. I'm only assuming Connor will have some knowledge to pass down as well, otherwise viewing his memories would be pointless.
3 messages delivered: Minerva, Juno, and Jupiter.

We know lucy lost her life and clay but who else

I think it has to be Juno, Minerva, and Jupiter. They toiled to save the planet, they failed and all died, they each delivered a message to Desmond.

With 3 lives lost, Lucy might not be counted, 'cos she's a templar!
3 lives lost - Clay, maybe Lucy, and, umm, can't think of one.... Mrs. MIles?
3 Lives toiled - Altair, Ezio and either Connor or Desmond
3 messages - Minerva, Juno and Jupiter's messages

Three lives lost can also relate to Adha, Cristina and Lucy, as they could be said to have been the first love/romances of all the Assassins (Altair, Ezio and Desmond) so far.
