Go to the Nvidia(or ATI) Control Panel. In 3D Settings set defaults and/or set to "Application Controlled".
Also another thing to try is setting your LCD Screen refresh rate to 60Hz. (You can find this in Display Settings, Advanced)
You may need to do the above two things along with turning Vertical Sync on.

Solution: Apparently caused by the game phoning home (gconnect.ubi.com), a possible solution is to disconnect from the Internet while playing the game although it's unclear whether or not this works for everyone.

No solution for this problem has been found at this point and the readme states that "Ubisoft will not be developing updates to fix possible problems that might occur on this type of computer."
------ Stuck in certain area due to game bug ------
If you're finding yourself stuck in a certain area where a certain event isn't triggering (doors that remains closed, areas that can't be healed after defeating the boss), a possible solution is to retrieve an older autosave. The game doesn't overwrite autosaves but only displays the latest one. On Vista, the list of save files can be found in "C:\Users\<your username>\Documents\Prince of Persia\Save". The autosaved games will have a filename such as "2008-12-17 22h53m46s Autosave.PoPSavedGame". If you want to use an older autosave, you simply have to remove (make a backup just in case) all the newer savegames. For those that want to increase the amount of saves available ingame, note that you can achieve this by making several copies of the non-auto saves (keep in mind I didn't thoroughly test this so don't blame me if you end up losing your savegames). This could work for console versions as well, I don't know if they also keep all of their autosaves.
